家蚕hop基因的克隆及序列分析 被引量:1

摘要 JAK激酶HOP是JAK/STAT信号途径的重要组成成员,在信号传导中发挥重要作用。为研究家蚕的JAK/STAT途径,通过RT-PCR克隆家蚕的hop基因(Bmhop)的cDNA,序列测定结果显示:Bmhop开放读码框为1 924 bp,编码638个氨基酸残基;结构分析结果显示:BmHOP具CRD_FZ、Kringle、转膜区域和PKc超家族蛋白激酶催化结构域TyrKc,每个结构域均可形成特定的三级结构;基于基因芯片的数据分析结果显示:Bmhop在家蚕5龄第3天的睾丸、卵巢、头部、脂肪体、中肠、马氏管中均有表达,但表达水平较低;基于TyrKc结构域序列的进化分析指出:昆虫HOP并不完全按物种聚类,BmHOP单独形成1个分枝,HOP在进化过程有TyrKc结构域扩增和TyrKc结构域组合事件发生。本研究结果为探讨BmHOP在JAK/STAT信号途径中的作用方式以及hop基因进化提供新的线索。
