家蚕SOCS-2基因的克隆与序列分析

摘要 SOCS基因是JAK/STAT信号通路的负调节基因。为了了解家蚕SOCS基因(BmSOCS)的遗传变异与进化,通过电子克隆获得了SOCS cDNA基因序列,结果显示家蚕基因组中至少存在4种BmSOCS基因,BmSOCS-2基因存在可变剪接。通过RT-PCR从家蚕中扩增得到1个BmSOCS-2基因(BmSOCS-2A),测序结果表明,该792 bp片段含有完整的ORF,由4个外显子组成,编码263个氨基酸残基,分子量为28.8 ku。结构分析显示,BmSOCS-2A有5个α螺旋结构,存在典型的SH2结构域和SOCS结构域。基因芯片数据分析显示,BmSOCS-2在家蚕雌雄性腺组织中表达量最高。进化分析表明,SOCS以不同种类聚类在一起,表明各种SOCS基因家族在各昆虫物种形成前就已产生。
