摘要
以实验室某长期稳定运行的UASB反应器的污泥为样品来源,通过降温筛选、分离纯化,得到一株在15℃条件下生长良好的产甲烷菌株US-3,在荧光显微镜下观察其菌落发出蓝色荧光,高倍扫描电镜观察菌体为较规则球状。经正交试验对其生长特性进行研究,试验结果表明,影响该产甲烷菌株生长能力的因素按主次依次为底物种类、温度、pH值、Na+浓度,各因素最佳水平为底物甲酸钠,温度25℃,pH值6.5,Na+浓度0.6mol.L-1。
A strain of methanogen which grew well at a low temperature of 15℃ was obtained from anaerobic sludge in a UASB reactor by selecting, separating and purifying. The colony emitted blue fluorescent when observed by fluorescence microscope. Their colonies were regular globe when observed by high-powered scanning electron microscope. The methanogen's growth charac- teristics were studied by an orthogonal experiment. The results showed that the influence of environmental factors on the methanogen's growth was sorts of substrate〉temperature〉pH value〉concentration of Na*, and the preferable levels of these factors were sodium formate used as substrate, temperature of 5 ℃, pH 6.5, and 6.5 mol Na+·L-1 accordingly.
