摘要
目的构建再生障碍性贫血(AA)实验动物模型,研究E838对AA模型小鼠造血机能的治疗作用。方法建模BALB/C小鼠经γ射线照射后尾静脉注射DBA小鼠脾脏细胞,药物治疗组口服E838,1次/d,共14 d,无菌饲养至第15天检测与AA相关的血常规、网织红细胞、骨髓细胞计数和病理学指标,判断E838对AA的治疗作用。结果模型组小鼠外周血常规指标、骨髓病理学等造血功能相关指标与AA临床表现基本相符;E838治疗组小鼠造血功能相关指标均较AA模型组小鼠有明显改善。结论成功构建了AA实验动物模型,E838对AA模型小鼠造血功能具有改善作用,其机制有待于进一步研究。
Purpose To study the therapeutic effect of E838 on hematopoietic function in aplastic anemia(AA) mice by using AA animal model. Methods BABL/C mice were used to set up the model by using whole body γ/-ray irradiation and caudal vein injection with thymic cells of DBA mice. Mice of the drug treatment group were given E838 daily for 14 days. At the fifteenth day, mice were sacrificed and the blood analysis, reticulocyte calculation, myelogram and some immunologic examination were performed. Results Mice of AA model group has marked reduction in the cells of three systems in peripheral blood, reticulocyte and marrow proliferation. The hematopoietic function of AA mice treated with E838 was recov- ered apparently. Conclusion AA animal model was established successfully. The E838 seems to have therapeutic effect on hematopoietic function in AA mice.
