Determination of Benzo(a) Pyrene in Smoked Barbecue Meat Products by GPC-HPLC-RP

摘要 采用MPS全自动样品净化系统中的GPC凝胶渗透色谱净化熏烧烤肉制品中的苯并(a)芘(BaP),并用高效液相色谱荧光法(HPLC-RP)测定BaP的含量。GPC条件:GPC凝胶渗透色谱柱,以环己烷-乙酸乙酯(体积比50:50)为流动相,流速4.7 mL/min,收集时间为15~21 min。HPLC条件:Diamonsil C18色谱柱(250 mm×4.6 mm,5μm);流动相:乙腈-水(体积比88:12),流速为1.0 mL/min;柱温:30℃;进样量:10μL;激发波长:384 nm;发射波长:406 nm。该方法线性关系良好,加标回收率为82.4%~91.3%,相对标准偏差小于5%,检出限为0.15μg/kg。方法简化了手工操作,有效地去除了熏烧烤肉制品中的色素及脂肪干扰,可应用于熏烧烤肉制品中BaP的测定。 A method for quantitative detection of benzo (a) pyrene in smoked barbecue meat products by GPC of Accu Prep MPS and HPLC-RP was established. GPC conditions: gel permeation chromatography (GPC) was selected; mobile phase was cyclohexane-ethyl acetate (50 : 50), flow rate was 4.7 mL/min, and collection for 15-21 rain. HPLC conditions:Diamonsil C^8(250 mm x 4.6 mm, 5 μ m), flow rate was 1.0 mL/min, mobile phase was acetonitrile-water (88 : 12), column temperature was 30%2, sampling volume was 10μL, excitation wavelength was 386 nm and emission wavelength was 406 nm. The results showed that the standard curve of BaP had good linearity, the recovery rate was 82.4%--91.3%, the relative standard deviation was less than 5%, and the detection limit was 0.15 μg/kg. In this method, the manual operation was simplified, and the interference of pigment and fat were apart from smoked barbecue meat products. The method may be applied to determine the BaP in smoked barbecue meat products.
