心肌肌钙蛋白ⅠR146W突变对CaN-NFAT3信号通路的影响

摘要 目的探讨心肌肌钙蛋白ⅠArg146Trp(cTnI R146W)基因突变在钙调神经磷酸酶(CaN)-活化T细胞核因子(NFAT)3信号通路中的作用。方法将大鼠胚胎心肌细胞分为转染含cTnI R146W突变的腺病毒(A)组、转染野生型cTnI基因的腺病毒(B)组、腺病毒空载体对照(C)组和空白对照(D)组,Western blot法分别检测胞浆CaN、胞核NFAT3蛋白表达。结果各组心肌细胞中均有CaN蛋白表达,但组间差异无统计学意义(P>0.05);A组NFAT3蛋白表达显著高于B、C、D组(P<0.05)。结论 CaN-NFAT3信号通路参与cTnⅠ R146W突变导致心肌肥大的过程。 Objective To investigate the effects of cardiac troponin-I Arg146Trp(cTnI R146W) gene mutation on the calcineurin(CaN)-nuclear factor of activated T cell(NFAT)3 signaling pathway.Methods The rat fetal cardiomyocytes were divided into four groups of A(transfected with adenovirus containing cTnI R146W mutation),B(transfected with adenovirus containing wild type cTnI gene),C(transfected with adenovirus vector) and D(blank controls).The protein expressions of CaN and NFAT3 were detected by Western blot.Results The CaN protein was expressed in each group,but there was no significant difference among them(P〈0.05).The protein expression of NFAT3 in group A was obviously higher than that in groups of B,C and D(P〈0.05).ConclusionThe CaN-NFAT3 signaling pathway participates in the development of cardiac hypertrophy induced by cTnI R146W mutation.
