胃癌组织中GC-C和GN mRNA的表达及临床意义

Expression and significance of guanylyl cyclase C and guanylin mRNA in human gastric cancer

摘要 目的探讨鸟苷酸环化酶C信使核糖核酸(GC-C mRNA)及其配体内生性肽类激素鸟苷素mRNA(GN mRNA)在胃癌组织中的表达及其临床意义。方法应用实时荧光定量PCR法检测GC-C和GN mRNA在60例胃癌和相应30例远癌胃黏膜组织中的表达。结果 GC-C和GNmRNA在胃癌组织中高表达(40.95±25.20和36.95±21.66),而正常胃黏膜组织中不表达。两者在肠型胃癌中的表达高于弥漫型(P<0.01),而与年龄、性别、病灶大小、临床病理分期、分化程度、浸润深度、淋巴结转移和Borrmann分型等因素无关(P>0.05)。两者在胃癌中的表达呈正相关(P<0.01)。结论 GC-C和GN的异常表达与胃黏膜癌变有关,可能参与肠型胃癌的发生发展。 Objective To investigate the expression and significance of guanylyl cyclase C mRNA(GC-C mRNA) and guanylin mRNA(GN mRNA) in human gastric cancer.Methods The expressions of GC-C mRNA and GN mRNA were detected with real time PCR in 60 gastric cancer samples(group A) and 30 matched tumor-adjacent normal gastric mucosa tissues(group B).ResultsThe expressions of GC-C mRNA and GN mRNA in group A were 40.95±25.20 and 36.95±21.66,repectively,which were not expressed in group B.The expressions of GC-C mRNA and GN mRNA were more in the intestinal-type gastric cancer than those in the diffuse-type(P〈0.01).The expressions of GC-C mRNA and GN mRNA were not correlated with age,sex,tumor size,clinical stages,tumor differentiation degree,lymph node metastasis and Borrmann typing(P〈0.05).The expression of GC-C mRNA was positively correlated with that of GN mRNA in gastric cancer(P〈0.01).Conclusion Abnormal expressions of GC-C mRNA and GN mRNA in human gastric mucosa may play an important role in the carcinogenesis,particulally for the intestinal-type gastric carcinoma.
