摘要
1974年,Green 等提出用赖氨酸加苏氨酸可能筛选得到积累赖氨酸、苏氨酸或甲硫氨酸的突变体。迄今已在玉米,大麦,胡萝卜等材料中得到成功。本实验的目的在于建立有效的芦笋筛选体系并得到对赖氨酸加苏氨酸具抗性的愈伤组织,为进一步得到天冬氨酸族某些氨基酸含量提高的芦笋突变体奠定基础。
Asparagus officinclis calli were induced from shoot of seedlings.After mutagenization, two lysine plus threonine resistant mutant lines (LTR2,LTR3) were obtained by selection- nonselection-reselection procedures with 2 mmol/l lysine plus threonine.LTR2 and LTR3 calli remained resistance to lysine plus threonine after being subcultured for 1 year,and both of them showed cross resistance to 1 mmol/l aminoethylcysteine.In resistant calli,the free lysine, methionine and an unknown amino acid were 1-10 times more than those in controls.
