摘要
目的研究彩虹明樱蛤多糖(MIP)对转染HBV的HepG2细胞表达功能的影响。方法以不同浓度(10^-3-104μg/m1)MIV作用于体外培养的HepG2.2.15细胞,采用MTT法、时问分辨免疫分析法(TRFIA)和ELISA法检测MIP对HepG2.2.15细胞的毒性作用及HBsAg、HBeAg、Pre-SlAg表达的影响,采用FQPCR检测MIP对HBVDNA复制的抑制作用。结果MIP在10^-2~10μg/ml浓度范围内对HepG2.2.1s细胞无毒性作用,TC50为4633μg/ml。MIP在10^-2~102μg/ml浓度范围内,对HBsAg、HBeAg、Pre-S1AgDA及HBVDNA均有一定抑制作用,相应的治疗指数分别为129、28、20和83。结论MIP具有一定的抗HBV活性。
Objective To investigate the effects ofMoerella iridescens polysaccharide (MIP) on Hepatitis B virus (HBV) transfected HepG2 cells. Methods HepG2.2.15 cells were treated with different concentrations of MIP (10 2-104μg/ml), and they were cultured in vitro. The MTT assay, time-resolved fluoreimmunoassay (TRFIA), and enzyme-linked immunosorbent assay (ELISA) were used to explore the cytotoxic effect of MIP on HepG2.2.15 cells and its effects on HBsAg, HBeAg, and Pre-SIAg secreted by HepG2.2.15 cells. The fluorescence quantitative PCR (FO,:PCR) was employed to evaluate the inhibitory effects of MIP on the duplication of HBV-DNA. Results The results indicated that MIP had no clear cytotoxic effect on HepG2.2.15 cells in the concentrations from 10-2μg/ml to 102μg/ml, and the TCs0 of MIP was 4,633μg/m-k MIP has Some inhibitory effects in the concentrations from 10-2μg/ml to 102μg/ml on HBsAg, HBeAg, Pre-SIAg, and HBV-DNA. The therapeutic indices (TI) of MIP on HBsAg, HBeAg, Pre-SIAg, and HBV-DNA were 129, 28, 20, and 83, respectively. Conclusions MIP has a certain dose- dependent inhibitory effect on HBsAg, HBeAg, Pre-S1Ag, and HBV-DNA in the culture supernatant fluid of HepG2.2.15 cells; there is a dose-effect relationship, which shows that MIP has some anti-HBV activities.
