青蒿琥酯纳米脂质体抗人肝癌HepG2细胞作用研究 被引量:4

摘要 目的:将青蒿琥酯纳米脂质体与青蒿琥酯原料药对比,研究其体外抗肝癌作用。方法:应用青蒿琥酯纳米脂质体和原料药作用HepG2细胞,MTT法检测对细胞的增殖抑制作用;用hoechst 33258染色法观察细胞凋亡的形态学改变。应用RT-PCR检测细胞凋亡因子caspas-3表达。结果:青蒿琥酯纳米脂质体及原料药抗肝癌作用呈浓度、时间依赖性;青蒿琥酯纳米脂质体组caspase-3mRNA表达高于原料药组。结论:青蒿琥酯纳米脂质体可抑制肝癌细胞增殖,诱导其凋亡,青蒿琥酯纳米脂质体体外抗肝癌作用效果比青蒿琥酯原料药好。 Objectives: To compare the anti-hepatoma in liver cancer of artesunate API(Active pharmaceutical ingredient) with artesunate nano-liposomes.Methods: HepG2 cells were treated with artesunate API and artesunate nano-liposomes.Cell proliferation inhibition was measured by methylthiazolyl tetrazolium(MTT) assay.The morphological changes of apoptosis were observed by Hoechst 33258 staining.And RT-PCR was used to detect the mRNA expressions of the apoptosis-related factor of caspase-3.Results: The results indicated that artesunate API and artesunate nano-liposomes could inhibit the proliferation of HepG2 cells in a dose and time dependent manner.The mRNA expressions of caspase-3 were up-regulated,and the artesunate nano-liposomes group was higher than the artesunate API group.Conclusions: Artesunate nano-liposomes can inhibit the proliferation and induce apoptosis of human hepatoma cells of HepG2,and it shows much stronger anti-cancer effect than that of artesunate API.
