STZ复制实验性2型糖尿病大鼠模型过程及经验总结 被引量:9

PREPARATION AND EXPERIENCE OF RAT MODEL OF TYPE 2 DIABETES INDUCED BY STREPTOZOTOCIN

摘要 目的:建立具有胰岛素抵抗、近似人2型糖尿病临床表现的SD大鼠模型。方法:30只SD大鼠随机分为A组(对照组)6只、B组[小剂量链脲佐菌(STZ)素即STZ组]12只、C组(高脂高糖膳食加小剂量STZ组)12只。B、C组大鼠分别在普通膳食和高脂高糖膳食喂养4周后小剂量STZ(30mg/kg)腹腔注射5d。结果:B组成模率为75%(8/12),C组成模率100%(12/12)。成模大鼠均表现多饮、多尿、毛色枯黄、精神萎靡、形体消瘦等症状,血糖值均高于16.7mmol/L。结论:普通膳食+小剂量STZ(30mg/kg)和高脂高糖膳食+小剂量STZ(30mg/kg)均能建立2型尿病SD大鼠模型,但高脂高糖膳食+小剂量STZ成模率更高。 Objective:To set up a SD rat model similar to human type 2 diabetes mellitus with insulin resistance.Methods:Thirty SD rats were randomized into three groups,A: normal group(n=6),B:low-dose streptozotocin(STZ,30 mg/kg)group(n=12),C: high-fat high-sugar diet and low-dose streptozotocin(STZ,30 mg/kg)group(n=12).Groups B and C were induced by an injection of low-dose STZ(30 mg/kg) after 4 weeks of normal chow feeding and high-fat high-sugar chow feeding respectively.Results:The diabetes morbidity was 75%(8/12) in group B,100%(12/12) in group C.All rat model had symptoms of polydipsia and polyuria,bad fur and mental state,and emaciation.Conclusion:Both normal chow feeding associated with low-dose STZ(30 mg/kg) and high-fat high-sugar chow feeding associated with low-dose STZ(30 mg/kg) can establish type 2 diabetes animal model,but high-fat high-sugar chow feeding associated with low-dose STZ(30 mg/kg) had a high morbidity.
