癌—睾丸抗原SSX-2基因在儿童急性淋巴细胞性白血病外周血单个核细胞中的表达 被引量:1

摘要 目的:探讨癌—睾丸抗原SSX-2基因在儿童急性淋巴细胞性白血病(ALL)外周血单个核细胞中的表达,为判断ALL预后提供理论依据。方法:采用逆转录—聚合酶链反应,对51例ALL患儿外周血单个核细胞进行SSX-2mRNA检测,并进行SSX-2基因表达与临床特征的相关性分析;以30例非肿瘤患儿为对照组。结果:ALL初治患儿SSX-2 mRNA表达率为34.78%,对照组无一例表达。ALL初治组表达率明显高于对照组(P<0.05),ALL初治组表达率高于ALL完全缓解组表达率7.14%(P<0.05)。SSX-2基因的表达与儿童ALL性别、年龄、初诊时外周血白细胞数、免疫分型以及临床危险分级无关(P>0.05)。结论:儿童ALL患者SSX-2mRNA表达率较高,SSX-2mRNA表达可能与儿童ALL的发生、发展有关;SSX-2基因检测对儿童ALL近期疗效的评估、指导临床治疗有一定意义。 Objective:To investigate the expression of SSX-2 gene and its relationship and clinical implication in children with acute lymphoblastic leukemia(ALL).Methods:Reverse transcription polymerase chain reaction(RT-PCR)was used to detect SSX-2 mRNA expression in peripheral blood mononuclear cells of 51 children with ALL and 30 children with no-malignant diseases served as contro1 group.Results:The rate of SSX-2 mRNA expression in children in newly diagnosed ALL was 34.78%,while no case was positive in control group,which was remarkably higher in newly diagnosed ALL group than control group(P0.05).Especially,the positive expression rate of SSX-2 was 34.78% in newly diagnosed ALL,whereas it was 7.14% in complete remission in children with ALL(P0.05).The expression of SSX-2 gene was not associated with clinical characteristics(gender,age,WBC,immunophenotype,clinical risk) in children with ALL(P0.05).Conclusion:There is a high expression of SSX-2 mRNA in children with ALL.The expression of SSX-2 gene might play an important role in the pathogenesis of children with ALL.Examination of SSX-2 mRNA might have clinical significance guiding to select adaptive therapy in children with ALL.
