摘要
目的建立人血清中去甲万古霉素的HPLC测定方法,用于去甲万古霉素血清药物浓度的临床监测。方法色谱柱为Shima-pak C18(150mm×4.6mm,5μm),流动相为甲醇∶磷酸二氢钾缓冲液(pH=3.2)=15∶85,检测波长为236nm,流速1mL·min-1,柱温为35℃,去甲万古霉素为内标。结果去甲万古霉素和万古霉素的保留时间分别为5.8min和7.1min;标准曲线回归方程分别为Y=(0.000 203 27)X-1.624 06,r=0.999 2;线性范围为2.0~100.0mg·L-1。结论该方法便捷、准确、灵敏,适用于去甲万古霉素的临床血清药物浓度监测以及人体药代动力学研究。
Objective To establish a HPLC method for the determination of norvancomycin concentration in human serum.Methods The Shima-pak C18 column(150 mm×4.6 mm,5 μm) was used with a mobile phase of methanol/potassium dihydrogen phosphate buffer(15∶85,pH 3.2).The detection wavelength,flow rate and column temperature were set at 236 nm,1 mL·min-1 and 35 ℃,respectively.Norvancomycin was used as the internal standard.Results The retention time of norvancomycin and vancomycin was 5.8 minutes and 7.1 minutes,respectively.The regression equation of norvancomycin was:Y=(0.000 203 27)X-1.624 06,r=0.999 2.The calibration curves obtained were linear in the range of 2.0~100.0 mg·L-1.Conclusion The HPLC method is convenient,sensitive,accurate and reproducible for determining the concentrations of norvancomycin in human serum and investigating its pharmacokinetics.
