吴茱萸碱治疗痛风的药效学研究

目的:从降血尿酸、抗炎、镇痛以及抑制大鼠足关节肿胀四个方面进行吴茱萸碱的药效学研究,探讨吴茱萸碱治疗痛风的作用。方法:采用大鼠灌胃次黄嘌呤(100mg/kg),再皮下注射氧嗪酸钾盐(20mg/kg)致尿酸增高模型,观察吴茱萸碱的降血尿酸作用;采用二甲苯致小鼠耳廓肿胀法,观察吴茱萸碱的抗炎作用;采用小鼠醋酸扭体法,观察吴茱萸碱的镇痛作用;采用踝关节注射尿酸钠致大鼠急性足肿胀模型,观察吴茱萸碱对足关节肿胀的抑制作用。结果:与模型组比较,吴茱萸碱中、低剂量组(18、9mg/kg)降血尿酸有显著性;各剂量组(10、20、40mg/kg)对小鼠耳肿胀的抑制有显著性;各剂量组(10、20、40mg/kg)对醋酸引起疼痛的抑制有显著性;各剂量组(9、18、36mg/kg)对关节足肿胀的抑制有显著性。结论:吴茱萸碱具有降血尿酸、抗炎、镇痛作用,能减轻模型大鼠足关节肿胀程度,可望成为治疗痛风的一种化学成分。

Objective：The tests of lowering serum uric acid、anti-inflammatory、analgesia and inhibiting acute tumefaction on the foot of rats were aimed to investigate the pharmacodynamics research of Evodiamine,so as to further explore the effect of Evodiamine treating gout.Method：The method of feeding hypoxanthine（100mg/kg） firstly and then injecting oxygen hydrochloride acid potassium salt（20mg/kg） subcutaneously to establish the model of hyperuricemia mice was taken to investigate the effect of Evodiamine lowering serum uric acid;.The method of xylene-induced tumefaction on the ear of mice was taken to investigate the effect of Evodiamine ＇s anti-inflammatory;The method of acetic acid-induced mouse writhing was taken to investigate the effect of Evodiamine＇s analgesia;The method of sodium injection given to form the model of acute tumefaction on the foot of rats was taken to investigate the effect of Evodiamine inhibiting foot tumefaction.Results：Contrasted with the model groups,,middle-dose and low-dose groups of Evodiamine（18,9mg/kg） have significantly lowering serum uric acid effects;each dose group of Evodiamine（10,20,40mg/kg） has significantly antiinflammation effects,each dose group of Evodiamin（10,20,40mg/kg） has significantly analgesia effects;each dose group of Evodiamine（9,18,36mg/kg）has significantly inhibiting rat actue foot tumefaction effects.Coclusion：Evodiamine was shown to possess potent hypouricemic effects、antiinflammatory and analgesia.It also can ease the degree of tumefaction on the foot of rats,The results of those tests can prove Evodiamine may be expected to be a kind of Chemical composition to treat gout.
