栀子对STZ诱导的2型糖尿病大鼠血糖及血脂的影响 被引量:12

Effect of Gardeniae Fructus on decreasing hyperglycaemia and hyperlipemia in diabetic rats models induced by streptozotocin

摘要 目的:研究栀子水提取物、环烯醚萜总苷、京尼平苷、西红花总苷对链脲佐菌素致2型糖尿病大鼠血糖、血脂的影响。方法:采用高糖高脂饮食+链脲佐菌素腹腔注射造成大鼠2型糖尿病模型,分别灌胃栀子水提取物组(2g/kg)、环烯醚萜苷组(0.03g/kg)、京尼平苷(0.003g/kg),西红花总苷(0.02g/kg,)、二甲双胍组(0.25 g/kg)。连续给药4周。观测:①一般状况,如毛发、体重、尿量等;②测定血糖;③测定血清胰岛素,计算胰岛素敏感指数;④测定总胆固醇、甘油三酯、高密度脂蛋白胆固醇、低密度脂蛋白胆固醇。结果:与模型组比较,栀子各给药组大鼠体重有所增加,大便成形,小便较多。其中西红花苷组大鼠体重增加明显,24h尿量明显减少(P<0.05)。各给药组血糖均有所下降,但差异不明显。栀子水提物组(P<0.05)和京尼平苷组(P<0.01)大鼠的空腹血清胰岛素相较于模型对照组大鼠有所下降,而栀子水提物组(P<0.05)、环烯醚萜苷组(P<0.O5)和京尼平苷组(P<0.01)大鼠的胰岛素敏感指数明显高于糖尿病模型组大鼠。栀子水提物、西红花苷(P<0.05)和京尼平苷(P<0.01)能明显降低高糖高脂大鼠的胆固醇水平,栀子水提物(P<0.05)、京尼平苷(P<0.05)和西红花苷(P<0.01)能明显降低高糖高脂大鼠的甘油三酯,栀子水提物和西红花苷能升高高密度脂蛋白胆固醇(P<0.05),西红花苷能降低低密度脂蛋白胆固醇(P<0.05)。结论:栀子各提取物能在一定程度上降低2型糖尿病大鼠的高血糖和高血脂,其中栀子水提物为其降糖降脂的有效部位,京尼平苷对降低血清胰岛素,提高胰岛素敏感指数和降低血脂都有明显疗效,而西红花苷降低血脂的效果较为突出。 Objective:Study the effect of water extract of gardenia,iridoid glycosides,geniposide,and saffron total glycosides on blood glucose and lipid of streptozotocin-induced diabetic rats.Methods:Setting up diabetic rats model by high-sugar and high-fat diet,and intraperitoneal injection of streptozotocin.Intragastric administration of the corresponding drugs respectively for 4 weeks.Respective dose is the water extract of Gardenia jasminoides group(2g/kg)、gardenia total glycosides group(0.03g/kg)、genipin group(0.003g/kg)、crocin group(0.02g/kg)、metformin group(0.25g/kg).Observe the following indicators:①general situation in rats,such as hair,weight,urine output,etc;②Determination of blood glucose;③Determination of serum insulin and insulin sensitivity index;④Determination of total cholesterol,triglycerides,high density lipoprotein cholesterol,low density lipoprotein cholesterol.Results:After 4 weeks of administration,the weigh of rats in gardenia medication groups increased a lot,formed stool,and urine more,compared with model group.After treatment,the blood glucose of gardenia water extract group,Iridoid glycosides group,and saffron total glycosides group were lower than treatment before(P0.05).After treatment,the blood glucose of treated groups was lower compared with treatment before,but there was no significant difference.Fasting serum insulin in gardenia water extract group(P0.05) and geniposide group(P0.01) was lower than model group.The insulin sensitivity index in gardenia water extract group(P0.05),iridoid glycosides group(P0.05) and geniposide group(P0.01) was significantly higher than model group.Gardenia water extract,saffron total glycosides(P0.05) and geniposide(P0.01) can significantly reduce cholesterol levels in high glucose and high fat rats.Gardenia water extract,geniposide(P0.05) and saffron total glycosides(P 0.01) can significantly reduce triglyceride levels in high glucose and high fat rats.Gardenia water extract and saffron total glycosides can increased high-density lipoprotein cholesterol(P0.05).Saffron total glycosides can reduce low-density lipoprotein cholesterol(P 0.05).Conclusion:The extract of gardenia can reduce hyperglycemia and high blood lipids of type 2 diabetic rats in certain extent.Gardenia water extract was the effective part of blood glucose-lowering and lipid-lowering.Geniposide has significant effects to reduce serum insulin and improve insulin sensitivity and reduce blood lipids.The effect of lowering blood lipids of saffron total glycosides was prominent.
