摘要
目的探讨白细胞介素3l与转化生长因子13,在博莱霉素(BLM)致肺纤维化小鼠各时期肺组织中的表达规律以及关系。方法取48只雄性昆明种小鼠,随机分为两组:对照组和实验组,分别给予生理盐水和博莱霉素(5mg/kg)一次性气管内灌注后,第7、14、21和28d处死小鼠,应用WesternBlotting法测定肺组织中白细胞介素3l蛋白与转化生长因子β1蛋白表达。结果1)实验组肺组织中白细胞介素31与转化生长因子13,蛋白表达水平均高于对照组(P〈0.05)。2)实验组中白细胞介素31蛋白表达从第7d开始高于对照组,第21d达到高峰,第28d降低。转化生长因子β1蛋白表达高峰在第7d,此后无明显下降,保持高水平表达。结论白细胞介素31与转化生长因子β1在肺纤维化形成过程中存在高表达,尤其白细胞介素31在介导中后期肺纤维化进程中发挥重要作用。转化生长因子β1除本身具有促纤维化功能之外,还可能刺激白细胞介素31的表达。
Objective To investigate the IL-31 and TGF-1 protein expressions in lung tissues of experimental mice with pulmonary fibrosis (PF) which was induced by bleomycin in various periods, and to study the effects of IL-31 and TGF-1 in pulmonary fibrosis. Methods Forty eight male mice were divided into two groups: control group and experimental group, poured respectively by saline and bleomycin solutions and killed on the 7th, 14th, 21th and 28th day. Use Western Blotting method to analyse the protein expression of TGF-1 and IL-31. Results 1)The expression of IL-31 and TGF-1 protein in lung tissues in experimental group were higher than control group(P〈0.05). 2) The protein of IL-31 in lung tissues in experimental group were higher than control group from the 7th day, peaked on the 21th day, lower on the 28th day. The protein of TGF-1 were peaked on the 7th day, Conclusion The expression of IL-31 and TGF-1 were high on different stages of pulmonary fibrosis. IL-31 plays an important role especially on the middle and late stages in pulmonary fibrosis.
