摘要 水稻害虫是影响吉林省水稻产量的主要限制因素,为培育抗虫转基因水稻新品种,本研究采用农杆菌介导的遗传转化法,将人工合成的Cry1C*基因导入吉林省主栽粳稻品种吉粳88和吉利518中,共获得410个独立转化株。通过对转基因植株PCR检测,阳性率为67.2%;ELISA检测结果表明:不同株系Bt毒蛋白含量变化较大,变幅为0.40~4.86μg/g鲜叶重;田间抗性鉴定结果表明:2个株系表现高度抗性,且农艺性状与原品种没有显著差异。这两个高抗转基因水稻株系的获得,为进一步培育适宜于吉林省抗虫转基因新品种奠定了基础。 Rice insects are the main restriction factor influencing the rice production in Jilin province. Inorder to breed insect-resistant japonica rice, Cry1C* gene encoding Bt protein was introduced, mediated with Agrobacterium tumefaciens, into two elite restorer line Jijing88 and Jili518 of Jilin province. Four hundred and ten independently transformed rice plants were obtained. PCR analysis displayed that frequency of posive plants was 67.2%. ELISA of Bt protein showed that the different transgenic lines had the different expression levels. The expression levels of Cry1C* protein were 0.40~4.86 μg/g of fresh leaf weigh; Field experiment showed that two homozygous lines were higly resistant to rice stem borers and rice leaf folders naturally infested without changes for agronomic trait of their parents. The two transgenic lines will play an important role in breeding the insect-resistant japonica rice of Jilin province.
