摘要
采用242对微卫星DNA标记引物分析了29份番茄自交系的遗传多态性,从中筛选出扩增带型清晰、稳定的34对引物,34对引物的平均等位基因数为4.53,平均多态信息量(PIC)为0.5141,累积鉴别力(CDP)达0.999999。利用其中的11对SSR引物对杂交种‘浙杂210’、母本‘01-199-1-1-0’及7个疑似父本进行了亲子鉴定。结果表明,11个分子标记的三联体累积非父排除概率(CPE1)达0.9999999;二联体累积非父排除概率(CPE2)达0.999984,‘01-230’的累计PI达5024.3946,RCP为0.9998,大于99.73%,鉴定出‘01-230’是杂交种‘浙杂210’的真实父本。说明所选34对引物可用于番茄亲权鉴定,其判断成功率和准确率较高,结果可靠。
The polymorphisms of 29 tomato inbred lines were investigated by 242 pairs of microsatellite primers, and 34 pairs of microsatellite primers with clearly amplified bands and good reproducibility were screened, the mean number of allele was 4.53, the mean polymorphism information content (PIC) was 0.514 1, the cumulative power of discrimination (CDP) was 0.999 999. 11 pairs of microsatellite primers from 34 ones were used for parentage identification of 9 materials. The analysis results showed that the triplet cumulate excluding probability of paternity (CPE1) reached 0.999 999 9, the diad cumulate excluding probability of paternity (CPE2) was 0.999 984, the cumulate PI of ‘01-230’ material was 5 024.394 6, the relative chance of paternity (RCP) was 0.999 8, which was more than 99.73%.The results of this study revealed that 34 pairs of microsatellite primers could be used in the parentage identification of tomato with high assignment rate and accuracy.
