贝甲类海产品中创伤弧菌检验方法的探讨 被引量:5

摘要 目的探讨贝甲类海产品中创伤弧菌的检验方法。方法选用mCPC、CC和TCBS培养基比较创伤弧菌在3种不同培养基上的生长率,同时应用常规的生化鉴定方法、API 20E生化试剂条和PCR-vvhA法对20株分离自牡蛎的可疑创伤弧菌进行鉴定,比较3种鉴定方法的符合率。结果 20株创伤弧菌菌株在mCPC、CC和TCBS 3种选择性培养基上的生长率(PR)分别为(0.19±0.28)、(0.34±0.43)、(0.44±0.64),差异有统计学意义(P<0.01)。mCPC、CC培养基对人工染菌样品中不同浓度的创伤弧菌的检出率差异均无统计学意义(P>0.05)。mCPC、CC培养基对31份天然污染的贝甲类样品中创伤弧菌的检出率均为22.6%(7/31)。常规生化鉴定、API 20E生化试剂条、PCR-vvhA法对20株分离自贝甲类海产品的创伤弧菌可疑菌株的检出率分别为95%(19/20)、95%(19/20)和100%(20/20),差异无统计学意义(P>0.05)。结论对贝甲类海产品中创伤弧菌的检验选用以CC和mCPC培养基为基础的传统培养方法,结合使用分子生物学筛选及生化系统鉴定,可大大提高检测效率。
