摘要
建立高效液相色谱(HPLC)检测黑米中花色苷类物质的方法.黑米粉经石油醚和乙酸乙酯去脂后,70%乙醇浸提、超声波处理、浓缩后,进行高效液相检测.该方法花色苷质量浓度在0.005 2~0.052mg/mL内具有良好的线性关系(R2=0.999 8),平均回收率为99.72%,RSD为0.91%,花色苷最低检测限为0.1ng/mL.该方法操作简便快速,精密度和稳定性好,可用于从黑色稻米种质资源中筛选高花色苷含量的黑米品种.
The paper proposed a method to detect anthocyanin contents in black rice with HPLC. The black rice flour was extracted with ethyl acetate and petroleum ether under the assistance of ultrasound treatment followed by concentration. Then, the extract was analyzed by high performance liquid chromatography. A good linear relationship determined by HPLC method was observed when anthocyanin concentration was in the range of 0. 005 2--0. 052 mg/mL (R2 =0. 999 8). The average recovery rate {or anthocyanin was 99. 72% with 0. 91% of relative standard deviations (RSD), and the lowest limitation of anthocyanin is 0.1 ng/mL. The method, which is simple, stable and accurate, can be used for screening black rice varieties with high anthocyanin contents from black rice germplasm resources.
