新疆红花黄色素的分离提取与纯化的研究 被引量:2

摘要 研究了新疆红花黄色素的分离与提取、纯化与鉴定等工艺条件。以羟基红花黄色素A的提取率为考查指标,采用单因素实验法研究了溶剂的种类、用量、提取温度、提取时间与提取次数等因素对红花黄色素提取率的影响。采用硅胶柱层析法对红花色素进行纯化。HPLC和UV对单因素处理的分析结果表明,处理样品与10%乙醇提取液的料液比为1:50(m/v)、提取温度为60℃、提取次数为2次、提取时间为50min为最优提取工艺条件。获得的红花黄色素提取液经冷冻干燥后,以乙醇:丙酮:水=3:2:1配比进行硅胶柱层析,分离获得了以羟基红花黄色素A为主的两种红花黄色素纯化产物。 Study on separation,extraction and purification and detection of safflor Yellow(SY) in Sinkiang Carthamus tinctorius L.Using the extracting rate of Hydroxysafflor Yellow A(HSYA) as main indicator,the conditions of extracting SY were discussed,including extraction solvent,extraction temperature,extraction time and the times of extraction.SY was isolated by silica gel column chromatography and analyzed by UV and HPLC.The optimum extracting procedure of SY was by 50min of 10% ethanol decoct for 50min and extracting for 2 times under 60℃.The freezing-dried SY was separated and purified by silica column chromatography(ethanol:acetone:water=3:2:1),the two extracts containing mainly HSYA was obtained.
