摘要
目的:建立测定大鼠皮肤组织中盐酸西替利嗪含量的方法。方法:采用高效液相色谱法,以盐酸羟嗪为内标,检测6只大鼠外用盐酸西替利嗪20mg·kg-14h后皮肤组织中的药物含量。流动相为乙腈-0.05mo·lL-1磷酸二氢铵水溶液(330:700,V/V),流速为1.0mL·min-1,检测波长为229nm,进样量为20μL。结果:盐酸西替利嗪检测浓度的线性范围为0.80~96.0μg·mL-1(r=0.9992),定量下限为0.320μg·mL-1,低、中、高浓度的提取回收率分别为74.0%、75.1%、82.7%,日内和日间RSD分别小于6.9%和8.1%,大鼠皮肤组织中药物平均含量为(81.7±2.44)μg·g-1。结论:本方法操作简单、准确、灵敏,可用于大鼠皮肤组织中盐酸西替利嗪的含量测定。
OBJECTIVE:To establish the method for the content determination of cetirizine hydrochloride in rat skin tissue.METHODS:HPLC method was adopted to determine the content of cetirizine hydrochloride in 6 rats 4 h after smearing cetirizine hydrochloride 20 mg·kg-1 for external use taking hydroxyzine hydrochloride as internal standard.The mobile phase was consisted of acetonitrile-0.05 mol·L-1 ammonium dihydric phosphate solution(330:700,V/V) at the flow rate of 1.0 mL·min-1.The detection wavelength was set at 229 nm and the injection volume was 20 μL.RESULTS:The linear range of cetirizine hydrochloride was 0.80~96.0 μg·mL-1(r=0.999 2).The quantitative limit was 0.320 μg·mL-1.The extration recovery rates of cetirizine hydrochloride in low,medium and high concentrations were 74.0%,75.1 and 82.7%,respectively.The RSDs of intra-day and inter-day were lower than 6.9% and 8.1%.The content of cetirizine hydrochloride in rat skin tissue was(81.7±2.44) μg·g-1.CONCLUSION:The method is simple,accurate and sensitive,which can be used for content determination of cetirizine hydrochloride in rat skin tissue.
