摘要
目的检测恶性胸腔积液来源树突状细胞(DCs)及其培养上清中干扰素-γ(IFN-γ)、白细胞介素-10(IL-10)的表达,评估对淋巴细胞的影响。方法收集15例肺癌患者的恶性胸腔积液,密度梯度离心后贴壁获取DCs前体,以粒细胞巨噬细胞集落刺激因子(GM-CSF)、IL-4和肿瘤坏死因子-α(TNF-α)诱导培养DCs,流式细胞仪分析表型,ELISA法检测DCs培养上清中IL-10和IFN-γ的浓度。结果体外诱导培养后获得了典型形态的DCs,高表达CD83、HLA-DR和CD1a,成熟DCs上清中IFN-γ浓度明显增高,而IL-10浓度显著降低。结论恶性胸腔积液来源的DCs可能促进Th1反应。
Objective To detect the levels of interleukin-10(IL-10) and interferon-γ(IFN-γ) in the supernatant and the dendritic cells(DCs) of malignant pleural effusion,to evaluate the potential effect of DCs on the lymphocytes.Methods Precursory DCs were obtained by adherence after density gradient centrifugation of malignant pleural effusion of 15 patients with lung cancer,and were cultured with IL-4,granulocyte macrophage colony-stimulating factor and tumor necrosis factor-α.The Phenotypes of DCs were analyzed by flow cytometry,and the levels of IL-10 and IFN-γ in supernatant were measured by ELISA. Results DCs of malignant pleural effusion had the typical morphology after extracorporeal cultivation,and showed the high expressions of CD83,HLA-DR and CD1a.The level of IFN-γ in the supernatant was significantly increased when DCs became mature,and the level of IL-10 in the supernatant was significantly decreased when DCs became mature. Conclusion DCs of malignant pleural effusion can promote Th1 response.
