瑞舒伐他汀对实验性动脉粥样硬化兔动脉平滑肌细胞骨桥蛋白表达的影响 被引量:5

摘要 目的探讨瑞舒伐他汀对动脉粥样硬化(AS)模型兔主动脉骨桥蛋白(OPN)的表达变化。方法复制AS兔模型,检测血清脂质成分;HE染色观察动脉的结构;用免疫组化和Western blot检测主动脉中OPN的表达。结果成功建立了AS兔模型;实验用兔在高脂喂养12周后血清中脂质相关指标有明显的增加(P<0.05);HE染色则显示了模型主动脉内膜增厚,有泡沫细胞生成并可见明显的AS斑块;免疫组化和Western blot结果表明主动脉平滑肌细胞的OPN表达显著上升(P<0.05),治疗后OPN表达降低,AS斑块减少。结论瑞舒伐他汀可能通过降低OPN的表达从而抑制AS的形成。 Objective To study the effect of rosuvastatain on expression of osteopontin(OPN) in the artery wall of atherosclerotic(AS) rabbits.Methods The rabbit model of AS was induced by a high-cholesterol diet.We observed the structure of arteries by hematoxylin-eosin(HE) staining.OPN expression was detected by Western blot and immunohistochemical methods.Results The AS model was established successfully.The expression of OPN in the artery of high-lipid diet group increased markedly compared with that of control group.After treated with rosuvastatain in 12 weeks,the expression of OPN decreased compared with that of high-lipid diet group.Meanwhile,the HE staining indicated the foam cells products increased.Conclusion Rosuvastatain may decrease the expression of OPN,overexpression of OPN plays an important role in the AS.
