摘要
目的探讨人直肠癌中ChemoCentryx趋化因子受体(CCX-CKR)和血管内皮生长因子-C(VEGF-C)的蛋白表达及意义。方法采用免疫组化法检测50例人正常直肠黏膜组织(距肿瘤2 cm以上)、癌组织和局域淋巴结中CCX-CKR和VEGF-C的蛋白表达,ELISA法检测其血清中CCL21的浓度,并以此分析其与临床病理特征之间的关系。结果直肠癌组织和转移淋巴结中VEGF-C于胞膜和胞质显色;CCX-CKR蛋白在癌组织细胞的胞膜和胞质呈棕黄色。直肠癌伴淋巴结转移组中CCX-CKR表达的阳性率为25.7%,VEGF-C表达的阳性率分别为85.7%,与无淋巴结转移组比较,CCX-CKR和VEGF-C的表达差异均有统计学意义(P<0.01)。CCX-CKR表达阳性患者血清中CCL21的浓度低于CCX-CKR表达阴性的患者(P<0.01)。结论 CCX-CKR可降低CCL21的浓度,CCX-CKR和VEGF-C在直肠癌中的表达呈负相关,CCX-CKR可能对直肠癌肿瘤的发生和转移有抑制作用。
Objective To investigate the expression of CCX-CKR and VEGF-C in human rectal tumors and analyze the clinical significance.Methods Immunohistochemistry was used to detect the protein expression of CCX-CKR and VEGF-C in normal rectal tissues,rectal tumors,and peritumoral-lymphocytic in paraffin slides of 50 cases with rectal tumors;and ELISA was used to detect the concentration of CCL21 in blood serum.Results The protein expression of CCX-CKR and VEGF-C were positive in the cell member and cytoplasm of rectal tumors.Compared with no metastasis of lymph nodes,the expression of CCX-CKR was 25.7%(P0.01) and VEGF-C was 85.7%(P0.01) in the group of rectal tumors with lymph nodes metastasis.The concentration of CCL21 in blood serum in CCX-CKR positive expression patients was lower than that of the CCX-CKR negative expression patients.Conclusion CCX-CKR can reduce the concentration of CCL21 in blood serum,these are no correlation of the expression of CCX-CKR and VEGF-C.The CCX-CKR can inhibit the proceeding and metastasis of rectal tumors.
