牛视网膜微血管内皮细胞的选择性培养方法 被引量:2

摘要 目的探讨牛视网膜微血管内皮细胞(BREC)的体外选择性培养方法。方法体外分离牛视网膜组织,结合选择性培养基的使用等方法选择性培养BREC,Von Willebrand因子免疫组化鉴定,观察其形态学特点并绘制生长曲线。结果选择性培养的BREC能够连续传代,且纯度达到98%以上,对Von Willebrand因子抗体染色阳性,BREC融合后呈铺路石样单层生长,有接触抑制。其生长曲线显示该细胞在传代后的第3~4天进入对数生长期,到第10~12天进入平台期。结论选择性培养方法的应用可获得较高纯度的BREC,简单且重复性好,无需额外除杂步骤。
