Ras相关核蛋白和缺氧诱导因子HIF-1α在胃癌组织中的表达及临床意义

摘要 目的探讨Ras相关核蛋白(Ran)和缺氧诱导因子-1α(HIF-1α)在胃癌浸润、转移过程中的机制及其相互作用。方法采用免疫组化PV-9000二步法检测70例胃癌组织和70例癌旁正常胃组织(距癌灶5 cm以上)中Ran和HIF-1α的表达。结果胃癌组织中Ran表达水平显著高于癌旁正常组织(P<0.05),胃癌分期越晚、浸润越深、伴淋巴结转移其表达水平越高,在低分化胃癌组织和高、中分化胃癌组织中其表达水平的差异无统计学意义。HIF-1α在癌旁组织中的表达均为阴性,胃癌组织中的阳性表达率为77.14%,显著高于癌旁组织,差异有统计学意义(P<0.05),且与胃癌临床分期、浸润深度、淋巴结转移有关(P<0.05),而与胃癌分化程度无关。Ran与HIF-1α呈正相关,差异有统计学意义(P<0.05)。结论 Ran与HIF-1α的表达率均随着胃癌临床分期、浸润深度和淋巴结转移的发展而增高,它们共同参与胃癌的发生、发展,可作为胃癌浸润和转移的重要判定指标。HIF-1α可能通过上调Ran的表达,促进胃癌细胞增殖而促进胃癌的浸润、转移。 Objective To investigate the expression of Ran and HIF-1α in gastric carcinoma(GC) and study the correlation between Ran and HIF-1α in order to explore their roles in invasion and metastasis of GC.Methods The expression of Ran and HIF-1α were detected by PV-9000 two-steps method in 70 patients with GC.Results The expression level of Ran protein in GC tissues was significantly higher than that in the adjacent normal tissues(P〈0.05).The later the depth of invasion,clinical stage,lymphatic metastasis,the higher rate was the positive expression of Ran.The difference of expression level between differentiated degree was not found.The expression of HIF-1α in paired para-carcinoma was negative,but in GC the positive expression rate was 77.14%.The expression level of HIF-1α protein in GC tissues was significantly higher than that in paired para-carcinoma(P〈0.05).The expression of HIF-1α was significantly associated with the depth of invasion,lymph node metastasis and clinical stage(P〈0.05),but it was not associated with the degree of differentiation.The Ran was correlated with the HIF-1α expression(P〈0.05).Conclusion The expression of Ran and HIF-1α increase with the development of clinical stage,depth of invasion and lymph node metastasis.Therefore,the detection of the two proteins can be used as biomarkers to predict metastasis and prognosis of these patients.HIF-1α may induce tumor proliferation and invasion by activating the transcription of the Ran.
