摘要 目的:探讨甘草苷(Liquiritin,LQ)对人膀胱癌细胞T24增殖和凋亡的影响及其可能作用机制。方法:实验分为对照组、LQ20、50、100μg/mL组。MTT观察LQ对T24细胞增殖的影响,Annexin V/PI染色,流式细胞术检测LQ对T24细胞细胞凋亡的诱导作用,Western blot检测Caspase 3蛋白表达。结果:与对照组比较,LQ20、50、100μg/mL在24、48、72h均对T24有明显的细胞增殖抑制作用,LQ作用72h后细胞凋亡率增高,Casepase3蛋白含量明显增高。结论:LQ能抑制T24细胞的增殖和诱导细胞凋亡,其机制与Csaepase3蛋白表达增高有关。 Objective:The present study focuses on the effects of Liquiritin(LQ) on proliferation and apoptosis of human bladder cancer cell line T24 and its potential mechanism. Methods: Four groups including control, LQ20μg/mL, LQ50μg/mL and LQ100μg/mL groups were set up in this study. Cell proliferation was analyzed by MTY assay. Cell apoptosis of T24 induced by LQ was evaluated by flow cytometry with Annexin V / PI staining. Expression of Caspase 3 was detected by western blot. Results: Compared with the control, after treatment of LQ of 20μg/mL,50μg/mL, 100μg/mL for 24h ,48h ,72h, T24 cell proliferation decreased, OD value decreased,meanwhile,the apoptotic cells and Caspase 3 protein expression increased after treatment with LQ for 72h. Conclusion:The above results indicated that: LQ inhibited T24 cell proliferation, and also induced apoptosis, and these effects might be contributed to the increased Caspase 3 protein induced by LQ.
