肺气虚模型大鼠肺、肾组织水液代谢相关性的机制研究 被引量:16

摘要 目的:观察肺气虚模型大鼠肺、肾组织AQP1表达及血中ET、IL-1β、TNF-α水平的变化,为机体水液代谢过程中肺肾相关理论提供依据。方法:用气管内注入脂多糖及熏香烟方法复制肺气虚大鼠模型,应用免疫组化、Westernblot方法、RT-PCR方法测定肺、肾组织AQP1蛋白及mRNA表达的变化,应用放免法测定血中、肺组织匀浆中ET、IL-1β及TNF-α含量。结果:与对照组比较,模型组肺组织AQP1mRNA及蛋白表达明显下降;肾组织AQP1mRNA及蛋白表达明显增加(P<0.01)。模型组血中ET、IL-1β及TNF-α含量明显升高(P<0.01)。结论:肺气虚模型大鼠肺组织AQP1表达下调;肾组织AQP1表达上调;血中及肺组织中ET、IL-1β、TNF-α含量明显升高,结果提示肺主"通调水道"功能之一可能与影响肺肾组织AQP1表达有关。 Objective :To observe the expressions of AQP1 in lung and kidney tissues and the contents of ET ,IL - 1β ,TNF - α in blood in Lung - Qi deficiency model rats. The aim was to provide experimental evidence for the traditional Chinese medical theory of "the kidney is related with the lung in charge of regulating the water passage". Methods:The rats of lung - Qi deficiency were simulated by injecting lipopolysaccharide into the trachea with cigarette smoking. Immunohistochemistry and Western blot analysis were used to observe the expression of AQP1 ; RT- PCR was applied to analyze the expression of AQPI mRNA in lung and kidney tissues. Blood sample was taken and the concentrations of ET, IL - 1β, TNF - α were measured by radioimmunoassay. Results:AQP1 expression in the lung tissue in the model group was obviously lower than that in the control group, and so was AQPI mRNA. AQP1 expression in the kidney tissue in the control group was greater than that in the control group, and so was AQPI mRNA (P 〈 0.01 ). ET, IL - 1β, TNF - α concentrations of blood in the model group were markedly higher than those in the control group (P 〈0.01 ). Conclusion:In the Lung- Qi deficiency model rats, AQP1 expression down regulated in the lung tissue. AQP1 expressions increased in the kidney tissue. Concentrations of ET, IL -1βand TNF -α increased obviously in blood. One of the substance basis of "lung is the upper source of water" may be the AQP1 retro - expression in the lung and kidney tissues.
