摘要
目的研究鼻息肉组织中环氧化酶-2(cyclooxygenase-2,COX-2)、核转录因子-κBp65(nuclear factor kappa B,NF-κBp65)的表达以及二者之间的表达关系。方法选取鼻息肉患者40例,鼻中隔偏曲患者20例。通过鼻内镜手术,取鼻息肉患者部分鼻息肉组织及鼻中隔偏曲手术患者部分正常下鼻甲黏膜组织。应用免疫组织化学SP法检测环氧化酶-2及核转录因子-κBp65在鼻息肉组织及下鼻甲组织中的阳性表达,结果应用SPSS 14.0统计软件包处理数据。结果 40例鼻息肉组织中环氧化酶-2的阳性表达率为75%,环氧化酶-2在鼻息肉组织中的表达显著高于下鼻甲对照组,且差异具有显著性(P<0.01)。40例鼻息肉组织中核转录因子-κBp65的阳性表达率为85%,核转录因子-κBp65在鼻息肉组织中的表达显著高于下鼻甲组,且差异具有显著性(P<0.01)。Spearman相关性分析显示环氧化酶-2和核转录因子-κBp65之间存在显著正相关性(r=0.817,P<0.01)。结论环氧化酶-2与核转录因子-κBp65在鼻息肉中表达均增强,环氧化酶-2与核转录因子-κBp65存在显著正相关性。对二者以及其上下游各种因子研究将有助于揭开鼻息肉发生、发展分子机制。
Objectives To detect the expression of cyclooxygenase-2 (COX-2) and nuclear factor-kappaBp65 (NF-κBp65) and their relationship in nasal polyps. Methods 40 cases of nasal polyps and 20 cases of dewiation of nasal septum were enrolled. The nasal polyp samples and normal nasal mucus from the inferior turbinate were taken during the functional endoscopic nasal surgery operation. With the methods of the SP technique of immunohistochemistry, COX-2 and NF-κBp65 expression were measured between 40 human nasal polyps tissues and 20 inferior turbinate mucosas tissues,Microscope was used to calculate the pigmentation of each segment subjectively and ascertain the positive cell expressive rate. All the results were disposed by the SPSS 14.0 statistic soft package. Results The positive expressions of COX-2 in 40 eases of nasal polyps are 75%. The expression of COX-2 was significantly higher in nasal polyps than of inferior turbinates (P〈 0.01). The positive expressions of NF-κBp65 in 40 cases of nasal polyps are 85%. The expression of NF-κBp65 was significantly higher in nasal polyps than inferior turbinates (P〈 0.01).There was a positive correlation between COX-2 and NF-rd3p65 expression in nasal polyps tissues by using spearman correlation analysis. (r=0.817, P〈 0.01). Conclusion The high expression of COX-2 and NF-r, Bp65 in nasal polyps may play an important role in the pathogenes of nasal polyps. There was a positive correlation between COX-2 and NF-κBp65 expression in nasal polyps tissues. Study should be performed to confirm this conclusion and elucidate the precise role of NF-κBp65 and COX-2 in the initiation and promotion of nasal polyps.
