摘要
目的观察脓毒症大鼠心肌组织高迁移率族蛋白B1(HMGB1)mRNA的变化。方法 60只大鼠采用盲肠结扎穿孔法(CLP)制作大鼠脓毒症模型,分为2 h、6 h、12 h、24 h、48 h、72 h组,每组各10只,分别在相应的时间点处死;对照组和假手术组大鼠各10只,于72 h处死。通过实时荧光定量PCR法检测各组大鼠心肌组织HMGB1 mRNA的相对含量。结果不同时间点处死脓毒症组大鼠的心肌组织HMGB1 mRNA相对对照组含量平均值为2.18±1.31,假手术组为0.92±0.14,脓毒症组与对照组间差异有统计学意义(P<0.01)。脓毒症组大鼠心肌组织HMGB1 mRNA表达在24 h明显升高,48 h达到高峰,72 h仍处于高水平,不同时间点的HMGB1 mRNA相对含量差异有统计学意义(F=183.4,P<0.01)。结论脓毒症大鼠心肌组织HMGB1 mRNA增加,有必要进一步研究其在脓毒症心肌抑制中的作用。
Objective To study the mRNA expressions of the high mobility group protein box-1(HMGB1)in the myocardium of rats with sepsis.Methods Sixty of Wistar rats were subjected to an operation of cecal ligation puncture(CLP)and then randomly divided into 2 h,6 h,12 h,24 h,48 h and 72 h groups;and set up a normal group and a sham group with 10 rats each.The mRNA expressions of HMGB1 in cardiac muscles were taken to assay through the technique of real time PCR after operation.Results The average expression of HMGB1 mRNA in cardiac muscle of the rat with sepsis was significantly higher than that in both the normal group and the sham group control at CLP 24 h,and peaked at CLP 48 h.There were statistical significances among groups(P > 0.05),indicating the time dependent increase of the expression of HMGB1 mRNA in cardiac muscle of rats with sepsis.Conclusions The expression of HMGB1 is increased in the cardiomyum in rats with sepsis,further study is needed to understand how HMGB1 and sepsis induce myocardial suppression.
