益气活血方药早期干预对盆腔炎大鼠模型的影响

Influence of early intervention of herbs of supplementing qi for activating blood circulation on pelvic inflammation in experimental model

目的研究益气活血方药对盆腔炎大鼠模型病理组织学及免疫学的影响。方法以Wistar雌性大鼠为实验对象,采用机械损伤加混合细菌法建立盆腔炎动物模型,观察各组大鼠组织学改变及血清C3、C4、TNF-α、IL-2的浓度变化。分组方法:空白组——正常饲养;假手术组——损伤内膜,正常饲养;模型组——损伤内膜并注射菌悬液,正常饲养;早期干预组——损伤内膜并注射菌悬液第2天灌胃给药;常规治疗组——损伤内膜并注射菌悬液第15天灌胃给药。给药方法:按体表面积公式计算药量,每日给药一次,药量为3.96 g/kg,均给药14天。指标检查方法:TNF-α、IL-2:ELISA法;C3、C4:免疫比浊法。结果肉眼观察:早期干预组子宫形态基本正常,结构较清晰,与周围组织未见明显粘连;光镜下观察:早期干预组子宫内膜腺体结构较对照组清晰,间质分布较均匀;其大鼠血清C3、C4、TNF-α值与模型组相比,均有极显著性差异(P<0.01),并与常规治疗组相比亦有显著性差异(P<0.05);早期干预组及常规治疗组的IL-2值与模型组相比,均无显著性差异(P>0.05)。结论益气活血方药早期干预可有效抑制炎性子宫组织水肿及内膜腺体扩张,促进受损组织与细胞的形态结构及其生理功能的恢复,但尚不能明确此方药对盆腔炎大鼠模型IL-2有直接影响。

Objective To study the influence of herbs of supplementing qi for activating blood circulation on pelvic inflammation in experimental rats model on the immunology and histopathology.Methods Female Wistar rats were used and the pelvic inflammation model was established with mechanic injury combined with mixed bacteria injection,and observe the changes of histopathology and serum C3,C4,TNF-α,IL-2 in the concentration.Grouping method：blank group（feed normally）,sham operation group（injury the endometrium,and feed normally）,model group（injury the endometrium,inject bacterial suspension and feed normally）,early intervention group（via intragastric administration on the second day after injurying the endometrium,and injecting bacterial suspension）,routine treatment group（via intragastric administration on the fifteenth day after injurying the endometrium,and injecting bacterial suspension）.Administration methods：Administer medicine once a day,and the dosage is 3.96 g/kg,which is calculated dosage with formula of body surface area.They are given the medicine for 14 days.Detection method of index：The methods of tumor necrosis factor alpha and Interleukin 2 are elisa,and methods of complement C3 and complement C4 are immune turbidimetry.Results Visual inspection： The uterine shapes were basically normal,and the structures were clear which didn＇t adhere to surrounding tissues obviously in the group of early intervention.Light microscope inspection：endometrial gland structures are clearer than control group,and interstice are more uniform distribution in the group of early intervention.The group of early intervention of rats of the value of serum C3,C4,TNF-α compared with model group has significant differences（P0.01）,and there are very significant differences compared with routine treatment group（P0.05）.However,the value of serum IL-2 of the groups between early intervention and routine treatment group compared with model group has no significant differences（P0.05）.Conclusion Early intervention of herbs of supplementing qi for activating blood circulation could control tissue edema of inflammatory uterus and dilate of endometrial glands efficiently,and promote the recovery of morphology and physiological function of damaged tissues and cells.Yet can not clear the direct effect of the herbs on pelvic inflammation in the rat model of IL-2.
