摘要
基于配位相互作用的毛细管电泳技术,利用Cu2+与氨基酸的配位作用,形成具有不同离子淌度的络合物,在50 mmol/L的CuSO4缓冲液中可实现中药天冬中氨基酸的有效分离。在最优条件下,被分离的氨基酸分别为:赖氨酸、组氨酸、精氨酸、甘氨酸、色氨酸、亮氨酸、缬氨酸、谷氨酸。该法无需样品衍生处理,快速、简便,灵敏度较高,值得推广。
A capillary electrophoretic technology based on coordination interaction was applied in separating amino acids in Radix Asparagi.By coordination interaction,copper ions and amino acids formed complex with different ion mobility.An electrophoresis buffer of 50 mmol/L CuSO4 was found to provide the effective separation.Under the optimized condition,glutamic acid,histidine,arginine,glycine,tryptophan,leucine,valine,and glutamic acid could be well separated.The proposed method offered a number of benefits including rapid,simple,high sensitivity and no sample derivative process.It was worth popularization.
