柱色谱法分离甘油磷脂酰胆碱的研究 被引量:2

摘要 通过大豆粉末磷脂水解获得甘油磷脂酰胆碱(GPC)混合物,然后从3种典型的离子交换树脂中筛选出D113树脂作为分离GPC的主要树脂,研究了不同pH的解吸液对柱色谱分离GPC的影响。研究表明,不同条件下各组分的洗脱曲线和GPC纯度具有较明显的变化,pH接近7的情况下,在整个解吸过程中GPC纯度均较理想。解吸液通过特定的阴离子交换树脂的进一步纯化,所得产品经过HPLC分析,GPC的干基含量为97.11%。 The mixture of glycerophosphocholine ( GPC ) was prepared from soybean powder lecithin by hydrolysis. Three representative resins were compared, and Dl13 was selected to be the primary adsorption resin to separate the GPC. Different pH of desorption solvent made different effect on elution curve and the purity of GPC. As pH approached to neutrality, the purity of product in the whole desorption process was ideal. The elution of D113 resin column was further purified with a strong basic resin , and the purity of GPC product was up to 97. 11% on dry basis by. HPLC analysis.
