摘要
目的:观察α1-肾上腺素受体抗体(简称α1-受体抗体)介导的糖尿病大鼠对肾小管转化生长因子-β1(TGF-β1)表达的影响,探讨其在肾小管间质病变发生发展中的作用。方法:Wistar大鼠64只,分为对照组和糖尿病组。对照组和糖尿病组又分为无介导组与抗α1-受体抗体介导组。用链脲佐菌素复制糖尿病模型。葡萄糖氧化酶法测定血糖,放射免疫法测定尿白蛋白,免疫组化法检测肾小管基质TGF-β1的表达。结果:成功构建糖尿病大鼠模型,α1-受体抗体介导的对照组和糖尿病组肾皮质远端小管均可见TGF-β1表达,且糖尿病组明显高于对照组(P<0.01)。结论:TGF-β1在α1-受体抗体介导大鼠表达增加,即TGF-β1的表达与自身免疫有关。
Objective:To investigate the effect of autoantibodies against α1-adrenergic receptor on the expression of Transforming growth factor-β1(TGF-β1) in diabetic wistar rats and its mechanism of tubulointerstitial lesions were studied.Method:64 Wistar rats were divided into blank control rat group and the diabetic rat group.Blank control rat group and diabetic rat group were divided into non-mediated group and the autoantibodies against α1-adrenergic receptor-mediated group.The experimental diabetic rats were yielded by injecting streptozotocin.Blood glucose was measured by glucose oxidase.Serum creatinine was measured by radioimmunoassay.The expression of TGF-β1 in tubular ground substance was exanmined by immunohistochemistry.Results:Diabetic rats were modelinged successively.The TGF-β1 was exanmined in renal cortex distal tubule in both blank control rat group and diabetic rat group mediated by autoantibodies against α1-adrenergic receptor and the TGF-β1 expression level in diabetic rat group was significantly higher than that in blank control rat group(P<0.01).Conclusion:The expression of TGF-β1 in diabetic rats which mediated by autoantibodies against α1-adrenergic receptor is increased than that in healthy rats.The expression of TGF-β1 is related to autoimmune.
