HPLC法测定小细胞肺癌患者依托泊苷血药浓度探讨

Determination of Etoposide Plasma Concentrations of Small Cell Lung Cancer Patients

目的建立高效液相色谱法测定小细胞肺癌患者血浆中依托泊苷血药浓度的方法。方法色谱柱为Waters Symmetry C18(150 mm×4.6 mm,5μm),流动相为甲醇-水(45∶55,V/V);流速为1 ml/min,检测波长为220 nm,进样量为20μl,柱温为30℃。结果样品中各成分峰分离良好,检出限为1.295 mg/L,依托泊苷的线性范围为2.59～51.8 mg/Lr,=0.999 7,平均回收率为95.5%±0.9%(n=5),RSD为5%。结论本法操作简便、准确度高,适用于医院的临床监测。

Objective To detect etoposide blood plasma concentrations of patients with small cell lung cancer by high performance liquid chromatography（HPLC）.Methods Waters Symmetry C18（150 mm×4.6 mm,5 μm） was used as column.The mobile phase was methanol-water（45∶55,V/V）.The wavelength was 220 nm.The injection volume was 20 μl,column temperature was 30 ℃.Results Capylus of the element of the sample separated well,detection limit was 1.295 mg/L,linear range of etoposide was 2.59~51.8 mg/L,r=0.999 7,average recovery was 95.5%±0.9%（n=5）,with RSD of 5%.Conclusion The method is simple,highly precise.It is suitable for clinic monitoring.
