摘要 目的观察瘦素(Leptin)对子宫颈癌Hela细胞生物学行为的影响,并探讨其机制。方法用不同浓度Leptin重组蛋白处理子宫颈癌Hela细胞,MTT法检测细胞增殖状态的变化;流式细胞仪检测瘤细胞周期变化及凋亡;RT-PCR检测C-MYC基因的表达;Western blot检测C-MYC蛋白质的表达。结果 Leptin明显促进子宫颈癌Hela细胞的增殖并抑制细胞的凋亡;Leptin激活子宫颈癌Hela细胞中C-MYC基因及其蛋白质的表达。结论 Leptin通过激活C-MYC的表达促进子宫颈癌Hela细胞的增殖并抑制其凋亡,其作用机制通过激活癌基因C-MYC而发挥效应。 Purpose To investigate the effect of Leptin on the biological behavior of Hela cells of cervical cancer and to elucidate its molecular mechanism.Methods Hela cells of cervical cancer were treated with different concentrations of Leptin protein.The cell proliferation was detected by MTT assay and the apoptosis was analyzed by flow cytometry.At the same times,real-time PCR and Western blot were used to detect the expression of C-MYC oncogene in Hela cells.Results Leptin increased significantly the growth of cervical cancer Hela cells in a dose-dependent and time-dependent manner.Flow cytometry revealed that Leptin inhibited the cell apoptosis.C-myc mRNA and protein expression in Hela cells were up-regulated significantly.Conclusions Leptin can significantly increase the growth of cervical cancer,and its mechanism might be related to the activation of the oncogene C-MYC.
