摘要
目的:观察牛膝多肽对局灶性脑缺血后大鼠坐骨神经功能损伤的影响,为其应用于临床提供实验依据。方法:制备局灶性脑缺血-大脑中动脉阻塞模型,血管阻塞再通后连续6 d进行尾静脉注射牛膝多肽。术后7、30 d进行足迹实验,观察牛膝多肽对局灶性脑缺血后坐骨神经功能下降的影响。结果:于大脑中动脉阻塞后7、30 d,SD大鼠坐骨神经功能指数的百分率明显增加(P<0.01)。尾静脉注射不同剂量的牛膝多肽后,坐骨神经功能指数百分率均有不同程度的下降,其中牛膝多肽中剂量组(ABPP(0.2 mg/kg)),无论是大脑中动脉阻塞后7 d还是30 d,其坐骨神经功能指数百分率均较生理盐水组明显下降(P<0.01)。结论:牛膝多肽对大脑中动脉阻塞引起的坐骨神经功能下降具有保护作用。
Objective: To observe the effects of A.bidentata polypeptides(ABPP) on the sciatic function in focal cerebral ischemic rats,and provide the experimental evidence for its clinical applications.Methods: Middle cerebral artery occlusion(MCAO) was employed to establish in rats,then ABPP was injected daily via the tail vein for 6 days.After 7 days and 30 days,the sciatic nerve function index was measured to observe the neuroprotective effect of ABPP.Results: 7 days and 30 days after MCAO,the percentage of sciatic nerve function index of rats increased significantly(P0.01).However,ABPP,especially in dose of 0.2 mg/kg,could inhibit the increased percentage of sciatic nerve function index induced by MCAO in rats(P0.01).Conclusion: ABPP have neuroprotective effect on sciatic nerve dysfunction induced by MCAO.
