摘要
目的 探讨高渗造影剂对甘油所致肾损害的大鼠肾毒性作用及可能机制.方法 32只SD大鼠随机分为正常对照组(N组)、甘油对照组(G组)、泛影葡胺对照组(LM组)、甘油+泛影葡胺组(G+LM组),每组8只.G组和G+LM组给予50%的甘油,后腿深部肌肉注射;N组和LM组以同样的方法给予等体积的生理盐水.3天后,LM组和G+LM组再给76%泛影葡胺(10 ml/kg)尾静脉注射;N组和G组按同样方法给予等量的生理盐水.各组于5天后检测血清肌酐(SCr)水平,苏木精-伊红染色观察肾组织病理损害,免疫组织化学法检测肾组织骨形成蛋白7(BMP-7)的表达.结果 BMP-7在N组肾组织中有高度表达,G组表达减少,LM组有中度表达,而G+LM组表达明显减少.结论 泛影葡胺能加重SD大鼠原有的急性肾毒性,骨形成蛋白7表达的下调是肾损伤的加重的重要因素之一.
Objective To explore the possible effect and mechanism of nephrotoxicity of high - osmolar which applys in renal injury that glycerol induced. Methods 32 SD rats were randomly divided into normal control group (group N) , glycerin control group ( group G) and lodipamide meglumine control group ( group LM ) , glycerin + lodipamide meglumine groups (G + LM) (n = 8/group). G and G + LM two groups, given 50% of glycerin levels by 10 ml/kg weight in hind - leg muscles injection, equal volume of normal saline ( 10 ml/kg) was injected through hind - leg muscles in rats from N and LM groups. Three days later, LM and G + LM groups was injected 76% of lodipamide meglumine ( 10 ml/kg) through tail vein and equal volume of normal saline ( 10 ml/kg) did like the former from N and G groups. Serum ereatinine (SCr) level was detected; The renal injury was assessed by hematoxylin and eosin (HE)staining; The expression of BMP -7 in renal tissues was detected by immunohistoehemistry assay. Results Expression of BMP -7 in the N group was in high level and that in the G group was less. The expression of BMP - 7 in the LM group was a little shorter than that in the N group, but the expression in G + LM group was significantly lower than other groups. Conclusion The nephrotoxicity of original acute renal injury induced by lodipamide meglumine is increased, and the expression of BMP - 7 is lower and that is one of the important reasons about increasing of renal injury.
