自絮凝产琥珀酸放线杆菌发酵产丁二酸过程中的细胞回用

摘要 为了解除丁二酸生产中的产物抑制作用,考察了沉降法收集自絮凝产琥珀酸放线杆菌YZ0819菌体,移除产物,加入新鲜培养基并继续进行丁二酸的发酵。结果表明经正常70g/L初始葡萄糖发酵结束后,收集细胞进行第二次发酵,在仅含有葡萄糖和水作为发酵培养基的情况下,细胞能消耗60g/L的葡萄糖,生成45.30g/L的丁二酸,丁二酸得率为75.50%。与正常发酵相比,在第二次发酵过程中虽然丁二酸的生产强度有较大幅度下降但整个过程几乎不产副产物甲酸,乙酸的量也略有下降,因此主产物丁二酸的量有所提高。通过对细胞的回用研究表明,将产物移除收集细胞进行回用,细胞仍具有较高的产酸能力,整个过程增加了丁二酸的总产量,降低丁二酸的生产成本。 In order to relieve inhibition function of products, spent cells of self-flocculating Actinobacillus succinogenes YZ0819 recovered via sedi- mentation was used in the second succinate fermentation. The results showed that, after the first batch fermentation with the initial concentration of 70g/L glucose, the collected spent cells could consume 60g/L glucose to produce 45.30g/L uccinic acid with the yield of 75.5%. Compared with the first fermentation, the content of some by-products such as formic and acetic acid were decreased, although the production intensity decreased deeply. The content of succinic acid was predominated in the fermentation liquor. The results showed that spent cell via first fermentation exhibited good production performance for succinic acid, and the total yield of succinic acid was enhanced. By this successive reuse method, the production of sue- cinic acid is cost-saving and comoetitive.
