摘要
目的观察原花青素(proanthocyanidin,PC)对氧化应激(oxidative stress)致大鼠心肌缺血再灌注损伤的保护作用。方法采用结扎大鼠冠状动脉左前降支(LAD)30min,复灌3h的方法,建立大鼠心肌缺血再灌注损伤模型,对照组、剂量组分别给予生理盐水、PC,阳性对照组给予银杏叶片(48mg/kg),观察PC对大鼠心肌梗死面积、心肌细胞凋亡及心肌酶学的影响。结果与假手术组比较,模型对照组大鼠的心肌梗死面积明显增大、心肌细胞凋亡指数与心肌酶学各指标均发生显著变化,差异均有统计学意义(P<0.01);与模型对照组比较,阳性对照组、PC剂量组降低了心肌缺血再灌注损伤模型大鼠的心梗面积、心肌细胞凋亡、减少心肌细胞磷酸肌酸激酶(CK)、乳酸脱氢酶(LDH)释放、心肌组织黄嘌呤氧化酶(XO)活性及血清丙二醛(MDA)含量,提高了血清一氧化氮(NO)水平、血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及谷胱甘肽(GSH)含量,差异有统计学意义(P<0.05或P<0.01),并存在剂量反应关系。结论 PC对心肌缺血再灌注损伤模型大鼠具有抗氧化应激作用,能够减少心肌细胞凋亡,从而减少心肌梗死面积,并存在剂量反应关系。
Objective To study proanthocyanidin on oxidative stress-induced myocardial ischemia-reperfusion injury in rats.Methods MIRI rat model was induced by ischemia for 30 minutes and then reperfused for 180 minutes.Control group and dose group were given normal saline and PC respectively.The positive control group was given Ginkgo(48mg/kg).Effect of PC on myocardial infarct size,myocardial apoptosis and myocardial enzyme was monitored.Results Comparing with the sham group,model control group rats significantly increased myocardial infarct size.Myocardial apoptosis index and serum creatine kinase of the indicators had significant changes(P0.01).Comparing with the model group,positive control group and PC-dose group reduced myocardial ischemia-reperfusion injury rat model of myocardial infarct size,myocardial apoptosis,decreased myocardial creatine kinase(CK),lactate dehydrogenase(LDH) release,myocardial tissue xanthineoxidase(XO) activity and serum malondialdehyde(MDA) content,increased serum nitric oxide(NO) levels,serumsuperoxide dismutase(SOD),glutathione had peroxidase(GSH-Px) activity and glutathione(GSH) content.The difference was statistically significant(P0.05 or P0.01),and there was a dose-response relationship.Conclusion PC on MIRI in rats with anti-oxidative stress can reduce myocardial apoptosis, thereby reduce myocardial infarct size.In addition,there is a dose-dependent relationship.
