藤梨根提取物对肺癌A549细胞裸鼠移植瘤的抑制作用 被引量:12

Inhibitory effect of Radix Actinidiae extract on transplanted tumors of the lung cancer cell line A549 in nude mice

摘要 目的研究藤梨根乙酸乙酯提取物对肺癌A549细胞裸鼠移植瘤的生长抑制及凋亡诱导作用,并探讨其作用机制。方法建立肺癌A549细胞裸鼠移植瘤模型,观察藤梨根乙酸乙酯提取物的作用,设置对照组、治疗组(低剂量组、高剂量组)。通过测量移植瘤体积变化绘制生长曲线,根据终末瘤质量比较计算抑制率,采用TUNEL法检测移植瘤凋亡指数,采用免疫组化法检测移植瘤Survivin蛋白表达、增殖指数,采用逆转录-聚合酶链式反应(RT-PCR)检测移植瘤Survivin mRNA变化。结果各治疗组经藤梨根乙酸乙酯提取物治疗后移植瘤生长缓慢,治疗结束时治疗组移植瘤质量及瘤体积明显低于对照组,高、低剂量组的瘤质量抑制率分别为69.00%和45.09%。各治疗组移植瘤组织凋亡指数则明显高于对照组,而Survivin mRNA及蛋白表达较对照组明显减少,高剂量组尤其明显,各组间上述指标比较差异均有统计学意义(P<0.01)。结论藤梨根乙酸乙酯提取物在体内对肺癌A549细胞生长有抑制作用,可以诱导移植瘤细胞发生凋亡,促进癌细胞凋亡并下调瘤细胞Survivin蛋白和mRNA表达。 Abstract: Objective To study the inhibitory effect and apoptosis-inducing action of acetidin extract of Radix Actinidiae on transplanted tumors of lung cancer A549 cells in nude mice, and to explore its mechanisms. Methods A model of transplanted tumors bearing lung cancer A549 cells was established in nude mice, and the effect of acetidin extract of Radix Actinidiae in vivo was detected. The control group and treatment groups (high-dose group and low-dose group) were set for comparison. According to the transplanted tumor volume, the transplanted tumor growth curve was drawn. According to the final tumor weight, the inhibitory ratio was calculated. Terminal deoxynucleotidyl transferase mediated UTP nick-end labeling (TUNEL) was used to determine the apoptosis index of transplanted tumors. Expressions of Survivin protein and mRNA in transplanted tumors were detected by immunohistochemistry (SP) and RT-PCR, respective- ly. Results The growth of transplanted tumors in treatment groups was slowed after taking the drug. The final tumor volume and tumor weight in treatment groups were smaller than those in the control group. Tumor weight inhibitory rates in the high-dose group and low-dose group were 69.00% and 45.09%, respectively. The apoptosis index of transplanted tumor in treatment groups was higher than that in the control group. Expression of Survivin mRNA and protein in treatment groups were lower than those in the control group, and they was more significantly decreased in the high-dose group. Differences were significant in the above indexes between each two groups ( P 〈 0.01 ). Conclusion The acetidin extrac of Radix Actinidiae has an inhibitory effect on growth of transplanted tumors of lung cancer A549 cells in nude mice, inhibiting proliferation activity of transplanted tumor, degrading expressions of Survivin protein and mRNA, and inducing apoptosis of cancer cells.
