慢性乙型肝炎病毒感染者外周血T辅助细胞1、2型因子的表达

T helper-1,2 phenotype cytokines expression in peripheral blood of the patients with chronic hepatitis Bvirus Infection

目的 了解慢性乙型肝炎病毒(HBV) 感染者外周血单个核细胞(PBMC) 中CD4 + T 细胞内T辅助细胞1、2 型(Th1/Th2) 因子、干扰素γ(IFNγ) 和白细胞介素4(IL4) 的表达情况,以测定Th0/Th1/Th2 细胞的百分数,探明Th1/Th2 细胞在慢性HBV 感染中的作用。方法 常规分离PBMC,在佛波二酯(PMA)、Ionomycin 、Monensin 的刺激下,采用流式细胞检测仪(FACS) 对慢性HBV感染者外周血单个CD4+ T 细胞内IL4 和IFNγ表达进行分析。结果 正常对照组,2 .3 % ～18 .6 %的CD4 + T 细胞为Th1 细胞,0 .9 % ～9 .2% 为Th2 细胞;在慢性HBV 感染者外周血单个CD4 + T 细胞中,以Th0 细胞居多;而Th1 细胞则随着慢性乙型肝炎肝脏炎症活动的加剧而明显增多,在炎症活动明显的慢性乙型肝炎,其Th1 百分数明显高于炎症活动呈静止状态的慢性乙型肝炎;Th2 细胞在慢性HBV 感染的不同阶段则较为恒定,但明显高于对照组。结论 Th1 细胞与炎症活动呈正相关,Th2 细胞可能与HBV

Objective The composition of T helper 0,1,2 cell(Th0/Th1/Th2) in chronic hepatitis B virve (HBV) infected individuals was investigated by determining the expression of IL 4/IFN γ in a single CD4 +T cell isolate from periphesal blood mononuclear cell (PBMC), and the role of Th1/Th2 cytokines in chronic HBV infection was analyzed. Methods PBMCs from chronic HBV infected individuals were separated routinely, stimulated by phorboe 12 myristate 13 asetate (PMA)/Ionomycin/Monensin, and the production of interleukin 4 (IL 4) interferon γ (INF γ) by CD4 +T cells in PBMC was determined by fluorescence activated call sorting (FACS) analysis. Results The percentage of IFN γ producing T cells and IL 4 producing T cells ranged from 2.3% to 18.6% and from 1.1% to 8.7% respectively in CD4 +T cells from non infected individuals. The majority of CD4 +T cells from PBMCs in chronic HBV infected individuals were Th0 cells. The percentage of Th1 cells increased significantly with the increasing of hepatic inflammation activity. The percentage of Th1 cells in advanced period of chronic hepatitis B was higher than that in stable period of chronic hepatitis B. The percentage of Th2 cells in CD4 +T cells from HBV infected individuals did not differ significantly in different stages, but higher than that of controls. Conclusions The results indicated that Th1 phenotype cytokines were associated with hepatic inflammatory activity of chronic hepatitis B and Th2 cells may be associated with the chronicity of HBV infection.
