摘要
为探讨瘢痕疙瘩成纤维细胞对干扰素α 2b(IFNα 2b)作用的反应性 ,用IFNα 2b分别处理体外培养正常皮肤与瘢痕疙瘩中的成纤维细胞 ,采用3H 脯氨酸掺入、胃蛋白酶消化法检测成纤维细胞胶原合成量。结果发现 ,瘢痕疙瘩成纤维细胞经103U/mL的IFNα 2b作用后的胶原合成量显著降低(P<0.01) ;但与正常皮肤成纤维细胞的反应不同 ,经104U/mLIFNα 2b作用后的胶原合成量却未进一步减少。这提示瘢痕疙瘩成纤维细胞对IFNα 2b下向调节胶原合成作用的反应敏感性在一定程度上有所降低。
To investigate the biologic behavior of keloid fibroblasts, human dermal fibroblasts cultured from normal skin and keloids were treated with IFN α 2b for 48h at 103U/mL and 104U/mL.Collagen production by fibroblasts was assessed by 3H proline incorporation into pepsin resistant, precipitated extracellular collagenous protein. The results showed that treatment with IFN α 2b at 103U/mL significantly reduced the collagen synthesis in keloid fibroblasts(P< 0.01). However,the collagen synthesis in keloid fibroblasts did not reduce further after exposure to IFN α 2b at 104U/mL.The results suggest that keloid fibroblasts characterized by the hyperproduction of collagen may be less sensitive to the inhibitory effects of IFN α 2b on collagen synthesis to a certain extent.
