摘要
目的 探讨天然中药槲皮素和姜黄素对脓毒症大鼠肝损伤的保护作用及其作用机制.方法 将大鼠随机分为正常对照组(A组)、模型组(B组)、槲皮素预防组(C组)、姜黄素预防组(D组)、姜黄素+槲皮素预防组(E组)及假手术组(F组).采用盲肠结扎穿孔(CLP)法制备脓毒症模型.造模后24 h处死大鼠,取腹主动脉血测定血清中转氨酶(ALT、AST)浓度,并取肝组织检测TNF-α及IL-6水平,同时行病理检查及荧光定量PCR测定其中PPAR-γ及NF-κB的表达.结果.①与A组相比,B组肝组织光镜下可见损伤性改变,药物预防组肝组织损伤性改变减轻;②血清ALT、AST水平B、C、D、E组明显上升(P〈0.05);且C、D、E组显著低于B组(P〈0.05);③B组大鼠肝脏组织中TNF-α、IL-6的水平显著升高(P〈0.05),且C、D、E组TNF-α、IL-6的水平明显降低(P〈0.05);④C、D、E组较B组能显著上调肝组织PPAR-γ的表达(P〈0.05),下调NF-κB的表达(P〈0.05),且药物联合预防组(E组)与药物单一预防组(C、D组)比较,差异有统计学意义(P〈0.05).结论.槲皮素和姜黄素通过上调PPAR-γ及下调NF-κB的表达,降低组织TNF-α、IL-6的水平,从而发挥脓毒症时对肝脏的保护作用,有效减轻肝脏炎症反应,且两者联合应用较单药效果更好.
Objective To investigate the protective effect of the natural traditional Chinese medicine - Quereetin and Curcum on the liver function in sepsis rats. Methods The rats were randomly divided into normal group (group A), sepsis group (group B), Quercetin group (group C), Curcum group ( group D) , Quercetin plus Curcum group ( group E), and sham - operated group ( group F). Cecal ligation and puncture(CLP) were used to make a sepsis animal too(tel. The rats in each group were sacrificed on 24 h after operation. Serum levels of ALT, AST were determined at the same time point. Tissues from liver were obtained to determine TNF - a, IL - 6 and observe the pathological changes ureter microscopy and used Real - time PCR to detect the mRNA expression of PPAR - γ and NF - κB. Results In group B, the hepatic functions and morphological structures were damaged; In group of medicine prevention groups, the damages were remited; Serum ALT and AST levels in groups B and medicine prevention groups were higher than those in group A and group F( P 〈0.05 ) ; Serum ALT and AST levels were significantly lower in group of medicine prevention groups than those in group B (P 〈 0. 05) ; In group B, the levels of TNF -α and IL -6 were obviously increased(P 〈 0.05) ; In group of medicine prevention groups, the levels of TNF - α and IL - 6 were obviously decreased ( P 〈 0.05 ) ; Compared with group B, the group of medicine prevention groups could up - regulated the expression of PPAR - γ and down - regulated the expression of NF - KB ( P 〈 0.05 ) ; There are significantly difference in group E and group C, D(P 〈 0.05). Conclusion Quercetin and Curcum can effectively protect the structure and function of liver through up - regulating the expression of PPAR - γ and down - regulating the expression of NF - KB to decrease the levels of TNF - a and IL - 6, alleviate Quercetin plus Curcum group is better than Quercetin group or Curcum group only.
