摘要
目的:观察TGF-β1对rhBMP-2促进成骨前体细胞(MC3T3-E1)增殖和分化能力的影响。方法:MC3T3-E1加入不同浓度的rhBMP-2和TGF-β1+rhBMP-2进行组织学观察,用MTT法检测细胞增殖,通过ELISA检测细胞ALP含量。结果:随着加入rhBMP-2浓度的增加,MC3T3-E1细胞的增殖逐渐增加,TGF-β1+rhBMP-2组增加更为显著(P<0.05)。TGF-β1+rhBMP-2组和rhBMP-2组在rhBMP-2浓度为25、50、100μg/L间存在统计学意义(P<0.05)。随着rhBMP-2浓度的增加,ALP值逐渐增加,TGF-β1+rhBMP-2组ALP值升高更为显著(P<0.01),TGF-β1+rhBMP-2组和rhBMP-2组在rhBMP-2浓度为25、50μg/L间有统计学意义(P<0.05)。结论:TGF-β1可促进rhBMP-2对MC3T3-E1的增殖和成骨分化作用。
Objective: The present study was aimed to study the effects of transforming growth factor-β1(TGF-β1) in osteoblast-like cell line MC3T3-E1 growth and differentiation after recombinant human bone morphogenetic protein-2(rhBMP-2) stimulating.Methods: MC3T3-E1 was grown in DMEM supplemented with 10%FBS.Logarithmic growth of MC3T3-E1 were divided into 3 groups as follows:(Group 1) DMEM medium(control),(Group 2) rhBMP-2 in different concentrations,and(Group 3) TGF-β1+ rhBMP-2 in different concentrations.Cell viability was measured by MTT assay.ALP activity was evaluated by Elisa test.We used analysis of variance(SPSS 16.0 for Windows software) to determine the differences observed among the groups;α=0.05 was considered significant.Results: The number of MC3T3-E1 cells increased with rhBMP-2 in a concentration-dependent manner.A significantly increased number of MC3T3-E1 cells was found at TGF-β1 + rhBMP-2 group(P0.05) in which also exhibited in a rhBMP-2 concentration-dependent manner.rhBMP-2 treated MC3T3-E1 cells had intensively enhanced ALP activity and expressed more significant in TGF-β1+ rhBMP-2 group(P0.01).Conclusion: The findings suggest that TGF-β1+rhBMP-2 or rhBMP-2 alone induce osteoblastic differentiation of MC3T3-E1 cell line.
