拉伸刺激对舌鳞癌细胞侵袭力及MMP-2表达的影响 被引量:1

摘要 目的:通过FX-4000柔性基底膜加载系统实现舌鳞癌Tca8113细胞的体外拉伸加载,观察拉伸刺激对Tca8113细胞侵袭力变化及基质金属蛋白酶-2(MMP-2)表达的影响,以探讨拉伸刺激在舌鳞癌侵袭转移中的作用。方法:选取2%拉伸幅度、频率2 Hz拉伸加载Tca8113细胞,以在相同培养小室中静态培养细胞作为对照。细胞在拉伸加载后行MMP-2免疫组织化学SP染色,用图像分析软件进行定量分析;Transwell小室肿瘤侵袭力实验测定Tca8113细胞侵袭力。结果:拉伸组细胞的侵袭力和MMP-2表达均高于对照组细胞,差异有统计学意义(P<0.05);不同拉伸时间下各拉伸组间MMP-2表达无统计学意义(P>0.05),侵袭力变化有统计学意义(P<0.05)。结论:拉伸刺激可以促进舌鳞癌细胞MMP-2的表达和侵袭力变化,在舌鳞癌侵袭转移中起着一定的作用。 Objective: To observe tensile stimulate expressing matrix metalloproteinases-2(MMP-2) and to investigate the effort of tensile stimulation in the invasion capacity and metastasis of Tca8113 cells in vitro.Methods: 2% 2 Hz strain tensile were loaded on the Tca8113 cells while unstrained cells were used as a control.The expression of MMP-2 was studied with immunohistochemical techniques and image software for quantitative analysis,the invasion capacity of Tca8113 cells was determined by Transwell in vitro.Results: The expression of MMP-2 and the invasion capacity of the strained group was obviously higher than the unstrained group(P0.05).The expression of MMP-2 didn't change as the straining time changed(P0.05),but the invasion capacity did(P0.05).Conclusion: Our results suggest that tensile stimulation enhanced the expression of MMP-2 in Tca8113 cell line and may be involved in the potential of tumor invasion and metastasis.
