青白散对慢性软组织损伤大鼠骨骼肌Ⅰ、Ⅲ型胶原蛋白表达的影响

Effect of Qingbai Powder on the Expression of Collagen-Ⅰand Collagen-Ⅲ in Skeletal Muscle of Chronic Soft Tissue Injury Rats

目的观察青白散对慢性软组织损伤大鼠骨骼肌中Ⅰ、Ⅲ型胶原蛋白表达的影响以及局部软组织的形态学改变。方法 54只雄性SD大鼠随机分为自然愈合组、云南白药组、青白散组,每组18只。采用慢性软组织损伤动物模型,分别予以生理盐水(10ml/kg)、云南白药(0.18g/kg)和青白散(0.54g/kg)灌胃,每日1次,连续14d。给药后第1周末、第2周末、第3周末,免疫组织化学方法检测大鼠慢性软组织损伤后骨骼肌中ⅠⅢ型胶原蛋白的表达,酶联免疫法检测骨骼肌中肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)含量的动态变化。结果青白散组肌肉再生速度、愈合质量显著优于自然愈合组,在不同时间点的骨骼肌中TNF-α和IL-6的含量均较自然愈合组显著降低(P<0.01),Ⅰ、Ⅲ型胶原蛋白的表达均较自然愈合组显著降低(P<0.01)。结论青白散能阻止胶原纤维的过度生成,降低致炎因子水平,从而有利于损伤组织的修复。

Objective To observe the effect of Qingbai Powder on the exp ression of collagen-Ⅰ and collagen-Ⅲ in skeletal muscle of chronic soft tissue injury（CSTI） rats.Methods Fifty-four male SD rats w ere randomized into natural ly healing group,Yunnan Baiyao group and Qingbai Powder group with 18 in eac h.The CSTI models were established.The naturally healing group,Yunnan Baiyao group and Qingbai Powder group were given 10ml/kg of normal saline,0.18g/kg of Yunnan Baiyao and 0.54g/kg of Qingbai Powder respectively through gastric feeding once daily for 14 days.At the first,second and third weekend after the administration,the expression of Collagen-Ⅰ and collagen-Ⅲ in skeleta l muscle of CSTI rats was detected by immunohistochemistry method,and the contents of tumor necrosis factor-α（TNF-α） and interleukin-6（IL-6） were detected by euzymelinked im muno sorbent assay.Results Comparing with the naturally hea ling group,the speed of muscle regeneration and healing quality in the Qingbai Powder group promoted significantly.In the Qingbai Powder group,the contents of TNF-α and IL-6 and the expression of collagen-Ⅰ and collagen-Ⅲ were significantly decre ased in comparing with those of the naturally healing group.Con clusion Qingbai Powder could inhibit the hyperplasy of collagen fibe rs,decrease the level of inflammatory factor and promote the reparation of inju ry issue.
