Selective Determination of Propafenone Enantiomers Level in Human Plasma by Reversed-Phase Liquid Chromatography

摘要 采用GITC柱前衍生化法同时分离和测定人血浆中(R)-PPF和(S)-PPF的浓度,在50~500ng·ml^(-1)浓度范围内有良好的线性关系;血浆中(S)-PPF的提取回收率、相对回收率、日内RSD、日间RSD分别为88.76%~91.45%、98.76%~101.22%、5.62%~6.79%、7.65%~9.01%;(R)-PPF的分别为88.98%~91.33%、98.31%~100.56%、5.02%~7.03%、7.88%~9.05%。 A reversed-phase liquid chromatqgraphy analysis with pre-coloum derivatization for determining the level of Propafenone enantiomers in human plasma was established. The standard curves were linear in the range of 50-500 ng/ml. The refined-recovery rate, relative recovery rate, within-day RSD and between-day RSD for (S)-PPF were 88.76 %-91.45 % , 98.76 %- 101.22 % , 5.62 %-6.79 % , and 7.65 %-9.01 % , respectively. Those for (R)-PPF were 88.98 % -91.33 %, 98.31 %-100.56 %, 5.02 %-7.03 %, and 7.88 %-9.05 %, respectively.
