枸杞多糖对糖基化终产物及视网膜神经细胞的影响 被引量:8

EFFECTS OF LYCIUM BARBARUM POLYSACCHARIDES ON THE FORMATION AND HARMFUL IMPACTS OF ADVANCED GLYCATION END PRODUCTS

摘要 目的探讨枸杞多糖(LBP)对糖基化终产物(AGEs)的形成及其致视网膜神经细胞损伤的影响。方法快速构建体外非酶糖基化系统,并以此为对照组,设立不同浓度的枸杞多糖干预组,于孵育过程中不同时间点取样,检测各组AGEs总含量、4-呋喃-2-糠酰-1-氢-咪唑(FFI)含量和果糖胺含量。构建兔视网膜神经细胞损伤模型,分别用不同浓度的枸杞多糖进行干预,检测各组细胞生长情况、增殖抑制情况和血管内皮生长因子受体2(VEGFR-2)及胰岛素样生长因子Ⅰ(IGF-1)的表达情况。结果枸杞多糖干预各组AGEs总含量和FFI含量较对照组明显降低(P<0.05),增长速率也明显低于对照组(P<0.05);果糖胺含量明显低于对照组(P<0.05),且能延迟峰浓度出现时间。枸杞多糖能明显下调VEGFR-2的表达(P<0.05)、抑制IGF-1的分泌(P<0.05)。结论枸杞多糖能够明显降低体外非酶糖基化产物的含量,拮抗AGEs对神经细胞的损伤作用,保护视网膜神经细胞。 Objective To study the impacts of Lycium barbarum polysaccharides(LBP) on the formation and harmful effects of advanced glycation end products(AGEs).Method AGEs was prepared and taken as control group.Five LBP groups at different concentrations were incubated at the same condition as the control group.The samples were taken at 0h,24h,48h,72h,96h,120h and stored under-20℃,then the AGEs content was detected under fluorescence spectrophotometer,and 4 furanyl-2-furanyl-1H-imidazole(FFI) and fructosamine(FMN) content under spectrophotometer.AGEs 20%(v/v) was used to induce the damage model in vitro.Three different concentrations of LBP were used as intervention according to the results of partⅠ.The growth and death of cells,and the expression of vascular endothelial growth factor receptor 2(VEGFR-2) and insulin-line growth fator-1(IGF-1) were observed.Results The AGEs and FFI content and the increase rate in all LBP groups were significantly lower than those in AGEs group(P0.05).The FMN contents in LBP groups were obviously lower than those in AGEs group(P0.05) and the time of peak concentration was significantly delayed compared with AGEs group.AGEs could inhibit the growth of cells and increase the expression of VEGF and IGF-1(P0.05).Conclusion: LBP could reduce the inhibiton of cells growth,VEGFR-2 and IGF-1express to lighten the harm of AGEs on cells.
